The duration of CR response in the first line has increased from 12 → 16.1 months.

For those who reached the over 6-month milestone, survival was noted (previously in the thread, below) to be longer, so it can be used as a good prognostic indicator. However, duration of response (DOR) in trials is usually reported as a median. From the Moffitt study:

The effect of aza alone on CR duration is difficult to find due to such heterogeneous studies, but OR (overall response) is generally considered to last 10.1 months.

In the Verona trial, a phase 3 that ended in a scene-shaking failure, CR percentages were 20% and 18% in the control and active treatment groups. CR duration wasn’t even reported there because the venetoclax group had a miserable 18% CR, and the dosage of aza and ven had to be reduced due to bone marrow issues.

In Verona 1b, the CR% was 30%. https://ashpublications.org/blood/article/145/11/1126/534406/Efficacy-and-safety-of-venetoclax-plus-azacitidine The CR duration was 16.6 months, one factor that caused hype and expectations. Ultimately, the combination’s bone marrow toxicity ended its run.

Bex+aza CR is now 45%. CR duration is now 16.1 months and no bone marrow toxicity has been observed like in Verona; in fact, preliminarily the opposite. Patient numbers are small, but it still looks good.
